What is the NF-κB pathway?

The NF-κB (nuclear factor kappa-light-chain-enhancer of activated B cells) pathway is a transcription-factor signalling cascade that regulates expression of genes involved in inflammation, immune response, cell survival, and cell proliferation. The published downstream mechanism for KPV anti-inflammatory pharmacology following PepT1-mediated intestinal-epithelial uptake.

What the research literature says

NF-κB is a family of transcription factors (RelA/p65, RelB, c-Rel, p50, p52) that exist as dimers held inactive in the cytoplasm by IκB inhibitor proteins. Activation by inflammatory stimuli triggers phosphorylation of IκB by the IKK kinase complex, leading to IκB degradation and release of active NF-κB dimers that translocate to the nucleus and drive transcription of pro-inflammatory genes.

KPV’s anti-inflammatory pharmacology operates through downstream NF-κB-pathway modulation following PepT1-mediated intestinal-epithelial uptake. The Dalmasso PepT1-uptake characterisation (PMID 18061177) and the Kannengiesser IBD-research work (PMID 18092346) established the framework. The Viennois colitis-associated cancer work characterised the integration with broader inflammatory-pathway pharmacology (PMID 27458604).

TB-500’s angiogenesis-research mechanism also involves Notch/NF-κB pathway engagement in limb-ischemia models per the Lv work (PMID 32945357), though through a different receptor-engagement pathway than KPV’s intestinal-uptake framework.
