Definition
Lyophilization, also called freeze-drying, is a low-temperature dehydration process. A peptide solution is frozen, vacuum is applied to drop the surrounding pressure below the triple point, and the frozen water sublimates directly to vapour without passing through a liquid phase. The resulting product is a dry, porous, typically white or off-white solid — the lyophilized form, sometimes called a lyophilisate or cake.
Water is the substrate for most peptide-degradation reactions including hydrolysis, deamidation, and aggregation. Removing water dramatically extends shelf life. Properly lyophilized peptide held under cool, dry, light-protected conditions in a sealed container under inert gas can show multi-year stability, with the exact shelf life depending on the peptide’s intrinsic structural stability, the lyophilisation cycle parameters, and the storage temperature and atmosphere.
How lyophilization shapes peptide research workflow
For research-peptide supply, lyophilization is the standard format because it allows a single manufactured batch to be aliquoted, shipped at ambient or refrigerated temperatures (rather than frozen cold-chain), and reconstituted by the end researcher with bacteriostatic water at the time of use. The reconstitution step is where the peptide moves from solid-stable storage into solution where the standard degradation pathways become accessible.
Post-reconstitution stability is shorter than lyophilized stability. Researchers typically aliquot reconstituted peptide into single-use volumes to minimise freeze-thaw cycling, which can induce aggregation and oxidation in many peptides. Amber-glass vials, inert-gas headspace (argon or nitrogen), and refrigerated or frozen storage are standard handling defaults.
