BPC-157

BPC-157 is a synthetic fifteen-amino-acid peptide whose sequence corresponds to a region of a larger protective protein originally identified in human gastric juice. Studied across preclinical tissue-repair, gastric protection, vascular signalling, and neural research. Not approved for therapeutic use by any major regulator.

Definition

BPC-157 — pentadecapeptide BPC-157, also designated by the early development code PL 14736 and the proprietary research name Bepecin — is a synthetic peptide of 15 amino acids, sequence Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val (GEPPPGKPADDAGLV in one-letter code). The full name Body Protection Compound 157 refers to the parent gastric protective protein from which the active fragment derives. Canonical chemical identifiers: CAS registry number 137525-51-0, PubChem CID 9941957, molecular formula C62H98N16O22, molecular weight approximately 1419.55 g/mol.

The compound has been studied in preclinical research since the early 1990s, primarily by the Sikiric-Seiwerth research group in Croatia, with cross-laboratory replication of core findings published by independent investigators (PMID 30915550). Comprehensive reviews of the wound-healing (PMID 34267654), gastrointestinal protection (PMID 38980576), pleiotropic activity (PMID 38675421), and musculoskeletal repair (PMID 40789979) literatures consolidate more than three decades of preclinical work.

How BPC-157 is studied in peptide research

Preclinical research on BPC-157 spans tendon, ligament, skeletal muscle, bone-defect, dermal, peripheral-nerve, vascular, gastric, intestinal, and neural models. The dominant mechanistic finding is activation of VEGFR2 in vascular endothelial cells with downstream Akt phosphorylation and endothelial nitric oxide synthase signalling (PMID 27847966). Growth-hormone-receptor up-regulation is documented separately in tendon-fibroblast culture (PMID 21030672). The combined picture supports a multi-axis mechanism model with angiogenic, migration, and downstream-transcriptional components.

Translation to human clinical use is the open question that runs through every recent review. No major regulatory authority — Health Canada, FDA, EMA, TGA — has cleared the compound for therapeutic use in humans or animals. The published human-cohort literature consists of small pilot studies including a 2024 interstitial cystitis pilot (PMID 39325560).
